Interleukin-7 (IL-7, previously known as lymphopoietin-1 and pre-B-cell growth factor) is a hematopoietic growth factor that stimulates early B and T cells. It can also co-stimulate the proliferation of mature T cells with other factors such as ConA (concanavalin A) and IL-2. IL-7 can induce the formation of lymphokine-activated killer (LAK) cells, it also plays a role in the development of cytotoxic T lymphocytes (CTL). IL-7 can stimulate the production of pro-inflammatory cytokines and enhance the tumoricidal activity of monocytes/macrophages. It is produced by thymic stromal cells, spleen cells and keratinocytes. Human and murine IL-7 show cross-species reactivity.

ISOkine™ recombinant human IL-7 is produced in the endosperm tissue of barley grain (Hordeum vulgare), that exhibits up to 50 times less protease activity than E.coli or mammalian cells. Barley seed is void of any human or animal viral contaminants that could jeopardize your cell culture.

Reconstitution
Note: Always centrifuge the vial before opening. It is recommended to reconstitute the lyophilized protein in sterile water to a concentration of no less than 100 µg/ml. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Stability
The lyophilized protein, though stable at room temperature for few weeks, is best stored at -20°C. Reconstituted protein should be used immediately or stored in working aliquots at -20°C. Avoid repeated freeze-thaw cycles.
Biological Activity
Each batch is tested for bioactivity and verified to have comparable activity to a commercial source. The bioactivity of human IL-7 was determined by dose-dependent cell proliferation assay on human bone marrow mononuclear cells, measuring intracellular ATP concentration with standardized luminometry. EC50 for this ISOkine IL-7 LOT lies in the range of 48-72 ng/ml, corresponding to 0,16 x10E6 U/mg specific activity. We recommend that the optimal concentration for each specific application be determined by an initial dose-response assay.

Molecular Weight
Recombinant human IL-7 contains 153 amino acids and a 16 a.a. histidine-based tag for a total length of 169 a.a. and has a predicted molecular mass of 19.7 kDa including his-tag. As a result of glycosylation, the recombinant protein migrates as two bands with an apparent molecular mass of 19 and 21 kDa in SDS-PAGE.
